Can anyone explain the process of creating a Turbografx CD game from Mp3 and ISO files?

Also, is there an emulator out there that can emulate Turbografx CD games without needed a physical CD, but using ISO files?

I've never actually used it, however I seem to recall (maybe incorrectly) reading something about Hu-Go supported ISOs... Of course this was a few years ago, and I have no idea as to what is Hu-Go's current status. It may however be a good place to start looking regardless.

You can look into Alcohol 120% and see if it supports what you need it to do for you.

You have to convert the mp3's into wav's, make a cuesheet (made by sega cuemaker or something else) and load it into a virtual drive, like alcohol or daemon-tools. Magic engine will support it fine, never tried hugo or something else on my own.
