Using Cassini, but can figure out how to get a game in ISO+Mp3 format to work. I already converted the only mp3 that came with the game into WAV and still I can't get the game working. There is no .cue file that came with the game.

Look at your Turbografx CD Emulation-Thread. Similiar question - same answer.
If you're not satisfied with the Sega Cue Maker, try the Cue Maker (for img, bin, mdf-files)from here:

But Sega Cue Maker should do it, specifically designed for Sega-CD and Saturn-Iso's.

I made a Cue file and still the game didn't work, Cassini kept giving a wrong cue format 2 error. Then, using Cassini, I converted the whole thing into a BIN/CUE and the game still wouldn't start up due to a "BIOS and lots of funky symbols" error. What's wrong?

Post your cue-file along with the names of the according files, you're using. Looks like your cue-file is somewhat wrong... wrong mode perhaps.
Load it in a virtual drive, like daemon-tools or alcohol and PM me of the results.
The probs with your TG16-CD were solved ?
